First one that comes to mind is Monkey Madness. I plunked down far too many resources and still haven't found the dentures. So that quest is a write-off.
Desert Treasure is another, but that is mainly because, even with broadband, I still lose My connection on Runescape every so often. I don't wish for this to happen when fighting one of those "bosses."
Icthlarin's Little Helper was a write-off, until I found a way to solve that puzzle in three moves. Otherwise it would've been a no-can-do, since I suck at those kind of puzzles (same with sliding ones as well).

I haven't had that much trouble on quests but one of the hardest was Summer's End, it took me a while to do the first room.

Mourning's End Part 2, and Desert Treasure.
Underground Pass can be hard, if you are below 60 like I was when I did it.

Mournings End Part 2. You constantly have to use prayer through out almost all of the quest for one

Although MM was relatively easy, it's the hardest quest I've done so far

mm is really only hard if u dont have 43 prayer, cuz the dungeon will kill u easily. Desert Treasure and Recipe for Disaster i guess make the top on my list, followed by Underground pass (kept falling

) and i guess Legends after that. Regicide aint hard less u got low agility

I would say Monkey Madness or Recipie For Disaster

Recipe for Disaster is the hardest quest because not only is it lengthy but you also have to do so many other quests to meet its requirements.
Recipe For Disaster:
Required Skills: 70 Cooking, 48 Agility, 20 Fire Making, and the ability to defeat a level 227 monster.
Required Quests: Cook's Assistant, Fishing Contest, Goblin Diplomacy, Big Chompy Bird Hunting, Biohazard, Demon Slayer, Murder Mystery, Nature Spirit, Priest in Peril, Restless Ghost, Witch's House, Gertrude's Cat, Shadow of the Storm, Lost City, Legend's Quest, Monkey Madness, Desert Treasure, Horror From the Deep. You must also have 175 Quest Points.
What You Need: Eye of Newt, Rotten Tomato, Greenman's Ale, 2 Lemons, an Orange, Pineapple, Cocktail Glass, Cocktail Shaker, Knife, Ashes, about 250gp, 2 pots of Flour, 3 buckets of Milk, 2 Eggs, 2 bowls of Water, 4 Asgarnian Ales, Ice Gloves, Charcoal, Spice (or Gnome Spice), a Fishing Bait, 2 loaves of Bread, Orange Slice, blue, green or purple Dye, 3 Bronze Wires, Needle, Fishbowl, Pestle and Mortar, Raw Cod, Cake Tin, a Kitten or Cat (any stage), Stews, Hatchet, Pickaxe, Ogre Bow, Ogre or Brutal Arrows, Iron Spit, Log, Tinderbox, Ogre bellows, Ball of Wool, Pot of Cream, Raw Sweetcorn, Hatchet, Machete, Pot, Raw Chicken, Dramen Staff, Anti-Dragonfire Shield, Rope, M'Speak Amulet, Ninja, Guard and Zombie Greegrees.

Regicide and Underground Pass were some of the easiest quests I've ever done. And I have a Quest Cape.
For me, MEP2. No discussion. The fail rate for the handholds is too stupidly high, the shades can be a pain, and the puzzle itself isn't that easy.
Summer's End was quite annoying too, although it was nowhere as long as Mourning's Ends Part II, so it gets eliminated.
Recipe for Disaster was very easy for me. Doing a few subquests here and there, and the Final Fight isn't hard either.
So yeah, for me, MEP2.

Ehh? No one has said Summers End yet? M.E.P. 2 was A LOT easier than I thought it would be, just very, VERY long.

Rfd was actually the hardest for me. The boss fight was extremely hard and it was really long.
Mourning's Ends 2 and Monkey Madness were close seconds. Ugh, MM was so hard.

MEP2 just took forever and was very challenging and annoying.

I like most quest people are saying.

I don't like to even start quest that don't have something I like, I haven't done summer's end but I hope I can.
